Course content
Potentiometry. Electrodes and cells. Glass pH-measuring electrode, experimental approach to measurement of pH. Ion-selective electrodes. Nikolsky-Eisenman equation. Electrodes with membranes. Experimental setups of potentiometric titration, evaluation of titration curves. Polarography/voltammetry. Basics of polarographic measurements. Mercury drop electrode and its properties. Sample preparation for polarographic measurement. Recent trends in voltammetry. Types of titration curves, their interpretation and applications. Electrogravimetry and coulometry. Conductometry and conductometric titrations. Basic approaches and instruments in optical emission spectrometry. Flame photometry. Atomic absorption spectrometry (AAS). Basic setup of atomic absorption spectrometer. Flame atomisation and eletrotermical atomisation. Calibration and applications of AAS. Molecular absorption spectrometry in UV/VIS range. Basic photometric quantities. Bouguert-Lambert-Beer law. Possibilities of identification of compounds. Quantitative analysis. Photometric (micro)titrations. Luminiscence spectrometry. Principles of infrared and Raman spectrometry. Nonspectrometric optical methods. Polarimetry, refractometry, interferometry. Nephelometry and turbidimetry. Extractions, extraction equilibria, definition of equilibrium constants. Extraction of solid compound by liquid and supercritical fluid. Extraction from liquid to liquid, extraction of neutral chelates, extraction of ionic associates. Solid phase extraction, extraction by gas. Chromatography. Basic principles, classification of chromatography according to the mechanism of separation, introduction of sample etc. Processing of chromatographic data. Qualitative and quantitative analysis. Gas chromatography. Liquid chromatography. Paper and thin-layer chromatography. Electromigration separation methods. Electrophoresis and isotachophoresis.

Learning outcomes
The aim of the course is to extend knowledge of classical analytical methods with basic knowledge about instrumental analytical methods.
Students after graduation of the subject "Analytical chemistry II" together with the subject C625 "Analytical chemistry II Laboratory course" are competent to execute the simpler procedures in analytical laboratories equipped with the basic accessories.

Current check of study - 2 written tests - 30 % of grade, written part of examination - 20 %, oral examination - 50 %;
