Course content
|
Basic principles of chromatography, classification of chromatographic techniques. Thermodynamic and kinetic aspects of chromatography, data processing, efficiency of separation. Quality of chromatographic separation, quantitative and qualitative analysis. Thin layer chromatography and paper chromatography. Basic setup and equations in column chromatography, mobile phase flow (carrier gas) through the column, principle of gas chromatography. Instrumentation of gas chromatography, preparation and selection of columns, classification of stationary phases, applications. Principle and instrumentation in high performance liquid chromatography (HPLC). Preparation of columns for HPLC, normal and reversed-phase systems. Separation of ionic compound using ion-exchange and ion-pair chromatography, influence of pH. Gradient elution and elution with programmed temperature. Size-exclusion chromatography. Bioafinity chromatography. Supercritical fluid chromatography (SFC). Chiral separations. Field flow fractionation (FFF), classification of electromigration separation techniques. Isotachophoresis. Capillary electrophoresis - principle, factors affecting migration, instrumentation, applications. Micellar electrokinetic chromatography, electrochromatography, application of electromigration techniques.

Learning outcomes
|
The aim of the course is an extension of basic knowledge in the field of chromatography and electromigration analytical separation techniques.
Graduate of the course is able to assess the usage of different analytical procedures and methods and critically evaluate achieved results. Graduate is aware of the fundamentals, instrumentation and the use of various chromatographic and electromigration techniques for analyzing substances depending on the size of molecules, polarity, or ion character.
