Course content
Safety and principles of work in microbiological laboratory. Nutrient media. Growth of microorganisms on culture media. Cultivation of microorganisms. Innoculation by various techniques. Microscopic examination methods - observation of living microorganisms in native preparation and dark field microscopy. Staining of capsules and spores. Gram staining. KOH test Giemsa staining. Acidoresistant staining Biochemical tests based on the conversion of carbohydrates and nitrogenous substances. Gram-positive aerobic and facultative anaerobic cocci Gram-negative cocci Gram-positive bacilli Gram-negative non-fermentig bacteria Gram-negative facultative anaerobic rods - Family. Enterobacteriaceae Gram-negative facultative anaerobic rods - Family. Pasteurellaceae Mycology - cultivation of clinically important fungi. types of culture media. Preparation and observation of native fungal preparation. Final laboratory exercise

Learning outcomes
The main goal of the laboratory practise is to acquaint students with principles and basic skills used in microbiological laboratories. It also includes information on work safety and good laboratory practice.
The student is able to orient in staining methods needed to determine the morphology of bacterial cells, preparation of nutrient media, culture techniques and tests used to identify microorganisms. Student also knows the basic genera of bacteria, morphology and arrangement of bacterial cells, the growth on culture media and the results of basic biochemical tests necessary to determine the unknown strain.
