Course content
Safety and principles of work in microbiological laboratory, aseptic work, microbiological laboratory equipment. Methods of sterilization and disinfection. Basic terminology. Nutrient media (liquid and solid) - preparation and sterilization. Inoculation of microorganisms, practice of various inoculation techniques, isolation of pure culture, preservation and recovery of bacterial cultures. Cultivation of aerobic, anaerobic, microaerophilic microorganisms. Growth of microorganisms on solid and liquid nutrient media - determination of macroscopic characteristics. Physical, chemical and biological factors influencing the growth and reproduction of microorganisms - pH, temperature, UV radiation, osmotic pressure. Microscopic techniques (bright and dark field microscopy, fluorescence m., electron m.). Staining techniques - Gram staining. KOH test. Negative staining. Staining of spores and capsules. Acid-fast staining. Giemsa staining. Viruses - laboratory diagnostics. Cultivation of fungi important in the field of human microbiology - cultivation media, microscopic techniques. Biochemical tests based on carbohydrate transformation, transformation of nitrogen compounds. Other biochemical tests - production of hydrogen sulphide, production of catalase, oxidase, utilization of citrate. Commercial biochemical tests.

Learning outcomes
Students are going to acquire the knowledge and skills to conduct various techniques used in laboratory of microbiology.
Students become acquainted with staining techniques, preparation of nutrient media and identification of microorganisms. Completion of the course is necessary for the further studies of special and clinical microbiology.

Written examination
