Course content
pH Measurement in different media, standards, standardization, buffers, pH a pK assignment, kinetic measurement (presentation of methods and instruments). UV-VIS Spectra measurement, spectral pK determination of weak acid, stopped-flow method of measuring of rapid reactions. (presentation of methods and instruments). Spectral observation of base catalyzed reaction in pseudo-first order conditions, influence of base catalyst concentration, influence of temperature, activation parameters estimation. Study of thermal behaviour of organic compounds using differential scanning calorimetry - determination of melting point, crystallization, and decomposition, or other temperature-depending physical transitions. Spectral determination of dissociation constant of model substrate. Infrared and Raman spectroscopy, measuring of model compounds, elaboration and interpretation of data obtained. Study of rapid reaction of model compound using stopped-flow technique. Analysis of unrecognized reaction mixture using GC/MS technique. 1H and 13C NMR spectroscopy, sample preparation, measuring of model compound spectrum, interpretation of spectrum of unrecognized chemical. Study of electrochemical properties of organic compounds using cyclic voltametry.

Learning outcomes
The main aim of the course unit is to introduce and practise main patterns of physical organic chemistry. Education proceeds in using of experimental techniques as spectral, kinetic, pH, and pK measurement, which are usually used for observation and study of physical and chemical properties of organic compounds. Students use instrumentation of institute, individually solve specific problems from the area of physical organic chemistry using model compounds.
After graduation, student can individually apply experimental techniques usually used for study of physical and chemical properties of organic compounds. He is able to offer experimental technique for solving particular problem, to set experiment, to realize measurement, and after this, he can analyze and interpret results of experiment.
Prerequisites
Fundamental knowledge of organic, physical, and analytical chemistry in the range of bachelor study.
