Předložená disertační práce se zabývá vztahy mezi enzymy lipidového metabolismu, mastnými kyselinami a progresí onemocnění diabetes mellitus 2. typu. Progrese diabetu byla určována úrovní hladiny glykovaného hemoglobinu, jakožto markeru dlouhodobé kompenzace glukózy v krvi.
Pro zjištění koncentrací mastných kyselin v plazmě byla použita kombinace ultracentrifugace a dvou chromatografických metod. První byla tenkovrstevná chromatografie, pomocí které byly lipidy rozděleny do jednotlivých tříd. Následně byl podíl jednotlivých mastných kyselin v těchto třídách po derivatizaci zjišťován na plynovém chromatografu Agilent Technologies 7890A GC System s plamenově ionizačním detektorem. Aktivita lipidických enzymů byla stanovena výpočtem jako indexy.
Po statistickém vyhodnocení výsledků bylo přistoupeno k vyhodnocení výsledků ve vztahu ke kompenzaci diabetu. Se změněným lipidovým metabolismem u tohoto onemocnění byly pozorovány i diference v profilu mastných kyselin v jednotlivých frakcích. Podrobnějšímu popisu těchto změn se věnuje následující práce.
Význam této práce podtrhuje fakt, že jen v České republice máme okolo jednoho milionu diabetiků, z nichž 90 % trpí právě diabetem 2. typu. Přínos ve zkoumání vlivu jednotlivých mastných kyselin a enzymů lipidového metabolismu dle mého názoru leží hlavně v efektivnějších dietetických doporučeních a udržení kompenzace diabetu u léčených pacientů.

This thesis deals with the relationship between enzymes of lipid metabolism, fatty acids and progression of type 2 diabetes mellitus. Progression of diabetes was determined by the level of glycosidic hemoglobin as a marker of long-term blood glucose level compensation.
The combination of ultracentrifugation and two chromatographic methods was used to determine plasma concentrations of fatty acids. The first thin layer chromatography, whose task was to divide the total lipids in each class, which were derivatized to processing gas chromatography using gas chromatograph from Agilent Technologies 7890 GC System with a flame ionisation detector. Lipid enzyme activity was determined by calculation as indices.
After a statistical evaluation of the results, the results were evaluated in relation to diabetes compensation. With altered lipid metabolism in this disease, the differences in the fatty acid profile in the individual fractions were also observed. A more detailed description of these changes is given below.
The importance of this work is underlined by the fact that only in the Czech Republic we have about one million diabetics, 90% of whom suffer from type 2 diabetes. The benefit in examining the effects of individual fatty acids and enzymes of lipid metabolism, in my opinion, lies mainly in more effective dietary recommendations and maintenance of diabetes compensation in treated patients.
