Diplomová práce se věnuje problematice stanovení Ag ve vzorcích práškových a obvazových materiálů OKCEL Ag-L, OKCEL Ag-T a kultivačních roztocích BHI a MALT metodou plamenové atomové absorpční spektrometrie.
Ověřována byla stabilita a homogenita vybraných materiálů a propracována technika úpravy vzorků. Pro rozklad práškových a obvazových vzorků byla provedena optimalizace mikrovlnného rozkladu. Za účelem kvantitativního převedení stříbra do analyzovaného roztoku byla k mineralizaci vzorků použita 65 % (m/m) HNO3 a 25 % (m/v) amoniak. Roztoky BHI a MALT byly před vlastní analýzou 25x ředěny. Za uvedených podmínek byl vliv matrice shledán jako nevýznamný a pro stanovení uvolněného stříbra do roztoku mohla být použita metoda přímé kalibrace. Detekční schopnost a citlivost navržené metody, charakterizovaná hodnotou detekčního limitu, respektive charakteristické koncentrace byla 0,2 mg/l a 0,081 mg/l.
Za účelem studia antimikrobiálních vlastností daných materiálů byl sledován vliv přítomnosti mikroorganismů Propionibacterium acnes, Aspergillus fumigatus, Candida albicans, Pseudomonas aeruginosa a Staphylococcus epidermidis na uvolňování stříbra do roztoku. Sledován byl dále vliv teploty, času a světla na uvolňování stříbra do roztoku.
Z naměřených hodnot vyplývá, že na uvolňování stříbra ze vzorku obvazu OKCEL Ag-T měl vliv mikroorganismus Pseudomonas aeruginosa. Na uvolňování stříbra ze vzorku prášku OKCEL Ag-L měl vliv mikroorganismus Aspergillus fumigatus po 2 dnech kultivace.

The thesis was aimed to the development of methodology for an accurate determination of silver in powdered and bandage samples of OKCEL Ag-L and OKCEL Ag-T and BHI and MALT culture medium by flame atomic absorption spectrometry method.
The method for sample preparation and optimization of microwave digestion, needful for characterization of investigated samples, was also developed. Using the specific sample treatment, including the simple 25-fold dilution of BHI and MALT culture medium with deionized water, and microwave digestion of OKCEL Ag-T and OKCEL Ag-L samples with nitric acid and ammonia, the determination was free of interferences. After that a simple calibration method could be adopted for attaining accurate results. Sensitivity and detection power of the proposed method given by 3 limit of detection and characteristic concentration, respectively were 0.2 mg.l-1 and 0.081 mg.l-1.
In order to study the microbial properties of all of the above mentioned materials, Ag was determined in BHI and MALT solutions in presence of several types of microorganism under variable experimental conditions. It was found that in the case of OKCEL Ag-T sample, the influence of microorganism onto releasing of silver to culture medium was found to be significant (p = 0,04). Concentrations of silver in culture medium were obtained for Pseudomonas aeruginosa, in comparison with other investigated microorganisms. For OKCEL Ag-L the influence of microorganism and time were found to significantly influence the release of Ag to culture medium. The highest concentrations were obtained for Aspergillus fumigatus after two days.
