Course content
Safety and principles of work in microbiological laboratory, aseptic work, microbiological laboratory equipment. Methods of sterilization and disinfection. Basic terminology. Nutrient media (liquid and solid) - preparation and sterilization. Inoculation of microorganisms, practice of various inoculation techniques, isolation of pure culture, preservation and recovery of bacterial cultures. Cultivation of aerobic, anaerobic, microaerophilic microorganisms. Growth of microorganisms on solid and liquid nutrient media - determination of macroscopic characteristics. Physical, chemical and biological factors influencing the growth and reproduction of microorganisms - pH, temperature, UV radiation, osmotic pressure. Microscopic techniques (bright and dark field microscopy, fluorescence m., electron m.). Staining techniques - Gram staining. KOH test. Negative staining. Staining of spores and capsules. Acid-fast staining. Giemsa staining. Identification of microorganisms using biochemical tests - biochemical tests based on carbohydrate transformations - oxidation, fermentation, VP and MR test; biochemical tests based on the conversion of nitrogenous substances - evidence of urease, decarboxylation of lysine, evidence of indole formation, reduction of nitrates to nitrites; other biochemical tests - presence of the enzyme catalase, cytochrome oxidase, sulfan production, citrate assimilation, bacterial motility test. Evaluation of biochemical tests and verification of hemolysin production (genus Staphylococcus, Streptococcus and other hemolytic microorganisms). Modern methods in microbiology (rapid screening tests, rapid methods, molecular biological methods - PCR; MALDI-TOF/MS). Mycology - direct observation of microscopic fungi in biological material, GT-test, production of chlamydospores and pseudomycelia in yeast, histological preparations.

Learning outcomes
Students are going to acquire the knowledge and skills to conduct various techniques used in laboratory of microbiology.
Students become acquainted with staining techniques, preparation of nutrient media and identification of microorganisms. Completion of the course is necessary for the further studies of special and clinical microbiology.
